目的 建立全臂丛神经损伤(total brachial plexus injury,TBPI)后对侧大脑皮层差异蛋白质谱,从分子水平研究TBPI后大脑运动皮层重组表现.方法 应用SD大鼠建立TBPI模型,随后应用同位素标记相对和绝对定量(isobaric tags for relative and absolute quantitation,iTRAQ)技术鉴定大鼠TBPI后1d大脑运动皮层的差异蛋白质.结果 共鉴定到1 293个蛋白质,其中差异表达蛋白质259个,包括214个上调的差异蛋白质和45个下调的差异蛋白质.其中有关大脑神经活动的差异蛋白质18个.差异蛋白质主要定位于细胞膜,发挥蛋白质定位、蛋白质转运、细胞内运输等生物过程,分子功能主要涉及离子结合与核苷酸结合.结论 TBPI后对侧大脑运动皮层有多种蛋白质表达异常,可能是损伤的急性期即出现了皮层重组的表现.“,”Objective To establish the protein spectrums of contralateral motor cortex differential proteins after total brachial plexus injury (TBPI) and investigate the performance of motor cortical remodeling at molecular level.Methods TBPI models were established in SD rats,and then the differential proteins of contralateral motor cortex after TBPI for one day using isobaric tags for relative and absolute quantitation (iTRAQ).Results Totally 1293 proteins were identified,including 259 differentially expressed proteins,214up-regulated and 45 down-regulated.18 differential proteins were related to brain development or axonogenesis.The differential proteins were mainly localized in plasma membrane,playing biological process of protein localization,protein transport and intracellular transport.The molecular functions of the differential proteins involved ion binding and nucleotide binding.Conclusion There are lots of differentially expressed proteins of contralateral motor cortex after TBPI,which may be the performance of motor cortical remodeling at molecular level after acute injuries.
