目的:观察槲皮素对人乳腺癌细胞系MCF-7增殖抑制作用及对Survivin基因表达的影响,探讨槲皮素抗肿瘤机制。方法:不同浓度的槲皮素处理MCF-7细胞后,采用MTT法检测MCF-7细胞增殖抑制作用并计算抑制率,流式细胞术分析槲皮素对MCF-7细胞凋亡的影响,RT-PCR检测MCF-7细胞中Survivin mRNA的表达水平,Western-blot检测Survivin蛋白表达变化。结果:槲皮素能够诱导MCF-7细胞凋亡,对人乳腺癌细胞有明显的抑制作用,且呈剂量时间依赖性,随着药物浓度增加,肿瘤细胞中Survivin mRNA及蛋白的表达水平均呈不同程度的降低。结论:槲皮素使乳腺癌MCF-7细胞停滞在G0/G1期而抑制细胞增殖,其机制可能是通过降低Survivin mRNA及蛋白的表达发挥抗瘤作用。
OBJECTIVE: To observe the effect of quercetin on the proliferation of human breast cancer cell line MCF-7 and its effect on the expression of Survivin gene, and to explore the antitumor mechanism of quercetin. Methods: After treated with different concentrations of quercetin for MCF-7 cells, MTT assay was used to detect the inhibitory effect of MCF-7 on proliferation of MCF-7 cells. Flow cytometry was used to analyze the effect of quercetin on the apoptosis of MCF-7 cells. The expression of Survivin mRNA in MCF-7 cells was detected by PCR and the expression of Survivin protein was detected by Western-blot. Results: Quercetin could induce the apoptosis of MCF-7 cells and inhibit the proliferation of human breast cancer cells in a time-and dose-dependent manner. With the increase of drug concentration, the expression of Survivin mRNA and protein in tumor cells Different degrees of reduction. CONCLUSION: Quercetin inhibits the proliferation of MCF-7 breast cancer cells in G0 / G1 phase by inhibiting the expression of Survivin mRNA and protein.
