目的:建立玉泉丸中葛根素含量的HPLC测定方法。方法:采用反相高效液相色谱法,使用AgilentHC-C18(250mm×4.6mm,5μm)色谱柱,以甲醇:水(30∶70)作为流动相。流速1.0mL/min,检测波长250nm。结果:葛根素在40~160μg/mL范围内线性较好(r=0.9999),平均回收率为99.71%(RSD为0.35%)。结论:该方法快速、简便、可靠、准确度高、重复性好,处方中其他成分对测定亦无干扰,分离效果好,可用于玉泉丸中葛根素含量测定。
Objective: To establish an HPLC method for determination of puerarin in Yuquan Pills. Methods: Reversed-phase high performance liquid chromatography (HPLC) was performed on an Agilent HC-C18 column (250 mm × 4.6 mm, 5 μm) using methanol: water (30:70) as the mobile phase. Flow rate 1.0mL / min, detection wavelength 250nm. Results: The calibration curve of puerarin was linear in the range of 40 ~ 160μg / mL (r = 0.9999) with an average recovery of 99.71% (RSD 0.35%). Conclusion: The method is rapid, simple, reliable, accurate and reproducible. The other components in the prescription have no interference with the determination, and the separation effect is good. The method can be used to determine the content of puerarin in Yuquan Pills.
