目的:研究表没食子儿茶素没食子酸脂(EGCG)对人前列腺癌细胞体外生长的抑制作用,以及其对细胞增殖周期的影响。方法:用不同浓度的EGCG处理体外生长的前列腺癌PC-3细胞,通过对细胞形态的对比观察和MTT法检测EGCG对PC-3细胞的抑制作用,通过流式细胞术检测其对细胞周期的影响。结果:与对照组相比,EGCG处理组可使细胞明显变圆,体积增大,细胞内颗粒及脱壁细胞增多;MTT法显示,EGCG能明显抑制前列腺癌PC-3细胞生长,并呈时间、剂量依赖性(P<0.05);流式细胞术显示,EGCG处理的PC-3细胞被阻止在G0/G1期,并有凋亡峰出现。结论:EGCG通过阻滞细胞的增殖周期对前列腺癌PC-3细胞体外生长有明显的抑制作用。
Objective: To study the inhibitory effect of epigallocatechin gallate (EGCG) on the growth of human prostate cancer cells in vitro and its effect on cell cycle. Methods: PC-3 cells were treated with different concentrations of EGCG in vitro. The inhibitory effects of EGCG on PC-3 cells were observed by MTT assay and cell cycle arrest by flow cytometry influences. Results: Compared with the control group, EGCG treatment group could significantly round the cells, the volume increased, intracellular granules and detached cells increased; MTT assay showed that EGCG can significantly inhibit the growth of PC-3 prostate cancer cells, and showed time (P <0.05). Flow cytometry showed that PCC-3 cells treated with EGCG were blocked in G0 / G1 phase and apoptotic peak appeared. Conclusion: EGCG can significantly inhibit the growth of PC-3 cells in vitro by blocking the cell cycle.
