[Objective] To induce roots of Scutellaria baicalensis Georgia by Agrobaaerium rhizogenes strains 1.2556 and optimize culture conditions of hairy root using response surface methodology. [Method] Hairy roots were induced by coculture. Based on single factor experiments, culture time, temperature, shaking speed, inoculation amount were analyzed by Box-Benhnken design and response surface methodology. [Result] Hairy root free grew on hormone MS medium after infection by Agrobaaerium rhizogenes strains 1.2556, which showed the representative characteristic of hairy root. The optimal culture conditions of hairy root were as follows: culture time 24.95 d, temperature 27.4 ℃, shaking speed 123.69 r/mim, inoculation amount 0.500 g. The average yield of fresh weight of hairy root (fm) reached 9.271 28 g per bottle under optimal conditions. [Conclusion] This study optimizes the culture conditions of hairy root, which can provide basis for further development and utilization of S. baicalensis.
