目的 :探讨细胞内游离 Ca2 + 在 As2 O3诱导的胰腺癌细胞株凋亡过程中的作用及 Fas、Fas配体 (Fas L )的变化和意义。方法 :2 μm ol/ L 的 As2 O3与胰腺癌细胞株 SW- 1990共同孵育后 ,H- E染色、光镜观察 ,抽提细胞 DNA进行琼脂糖凝胶电泳 ,上流式细胞仪检测细胞凋亡特征 ;用 Fura- 2荧光负荷技术测细胞内游离 Ca2 + 浓度 ([Ca2 + ]i) ,流式细胞仪检测 Fas、Fas L的表达情况。 结果 :2μmol/ L As2 O3作用于胰腺癌细胞 48h后 ,胰腺癌细胞出现典型的凋亡特征性改变 ,且凋亡过程中[Ca2 + ]i明显升高 ,Fas、Fas L 表达呈先升后降改变。 结论 :As2 O3在诱导胰腺癌细胞凋亡过程中 ,肿瘤细胞凋亡与 [Ca2 + ]i升高和 Fas、Fas L 表达有关。
Objective: To explore the role of intracellular free Ca2 + in As2O3-induced pancreatic cancer cell apoptosis and the changes and significance of Fas and Fas ligand (Fas L). METHODS: 2 μm ol/L As2O3 was incubated with pancreatic cancer cell line SW-1990, stained with H-E and observed by light microscopy. The DNA of the cells was extracted by agarose gel electrophoresis and cell apoptosis was measured by flow cytometry. Characteristics; The intracellular free Ca2+ concentration ([Ca2+]i) was measured by Fura-2 fluorescence loading technique, and the expression of Fas and FasL was detected by flow cytometry. Results: When pancreatic cancer cells were treated with 2μmol / L As2O3 for 48h, the pancreatic cancer cells showed typical changes in apoptosis, and the [Ca2 +] i increased significantly during apoptosis. The expression of Fas and Fas L was first increased. Change down. Conclusion : As2O3 induces apoptosis of pancreatic cancer cells. The apoptosis of tumor cells is related to the increase of [Ca2 +]i and the expression of Fas and Fas L.
