目的 探讨己酮可可碱(pentoxifylline,PTX)对人α1(Ⅰ)前胶原(COL1A1)基因启动子活性的作用及其机制。方法 构建含人COL1A1基因启动于序列-2483-+42 hp片段与氯霉素乙酰基转移酶(CAT)报告基因的重组体pCOLH2.5,将其瞬时转染至人皮肤成纤维细胞,加入PTX或/和胰岛素样生长因子-1(IGF-1)及胰岛素,测定CAT活性。结果 0.4mmol/L、2mmol/L和10mmol/L的PTX使pCOLH2.5的活性降至对照的(82±9)%、(58±8)%与(32±13)%。IGF-1与胰岛素能促进pCOLH2.5的活性。PTX能拮抗IGF-1与胰岛素的这一促进作用。结论 PTX能下调人COL1A1启动子的活性,而IGF-1与胰岛素能上调该启动子的活性。PTX能拮抗IGF-1与胰岛素的这一上调作用。
Objective To investigate the effect of pentoxifylline (PTX) on promoter activity of human α1 (Ⅰ) procollagen (COL1A1) gene and its mechanism. Methods Recombinant pCOLH2.5 containing human COL1A1 gene promoter sequence of -2483- + 42 hp and chloramphenicol acetyltransferase (CAT) reporter gene was constructed and transiently transfected into human skin fibroblasts. After addition of PTX or / And insulin-like growth factor-1 (IGF-1) and insulin, CAT activity was measured. Results 0.4mmol / L, 2mmol / L and 10mmol / L of PTX decreased the activity of pCOLH2.5 to (82 ± 9)%, (58 ± 8)% and (32 ± 13)% of the control. IGF-1 and insulin can promote the activity of pCOLH2.5. PTX antagonizes this promoting effect of IGF-1 and insulin. Conclusion PTX can down-regulate the activity of human COL1A1 promoter, while IGF-1 and insulin up-regulate the activity of this promoter. PTX antagonizes this upregulation of IGF-1 and insulin.
