目的 :研究异丙酚对鼠肝线粒体结构与功能的影响 ,以探讨其对肝毒性的影响。方法 :用定磷法测定了三种浓度 (2 5、5 0和 75 μmol)的异丙酚对鼠肝线粒体H+ ATPase水解活性的影响 ;荧光探剂DPH标记线粒体内膜 ,用日立 85 0荧光分光光度计测定异丙酚对鼠肝线粒体内膜流动性的影响。结果 :异丙酚 2 5 μmol对肝线粒体无明显影响 ,但 5 0、75 μmol可使线粒体H+ ATPase水解活性明显高于对照组 ,膜流动性也明显增加。结论 :异丙酚中、高浓度可使线粒体耗氧量增加 ,能量合成降低 ,线粒体内膜稳定性降低 ,肝损害因素增加
Objective: To study the effect of propofol on the structure and function of rat liver mitochondria in order to investigate its effect on hepatotoxicity. Methods: The effects of three concentrations of propofol (2, 5, 50 and 75 μmol) on the mitochondrial H + ATPase hydrolysis activity of rat liver were determined by the method of fixed phosphorus. Fluorescence DPH labeled mitochondrial inner membrane, Effect of Propofol on the Mitochondrial Endometrial Fluid Flow in Rat Liver by Spectrophotometer. RESULTS: Propofol 25 μmol had no effect on hepatic mitochondria but mitochondrial H + ATPase hydrolyzate activity was significantly higher than that of control at 50 and 75 μmol, and membrane fluidity was also significantly increased. Conclusion: Propofol at high concentration can increase mitochondria oxygen consumption, reduced energy synthesis, mitochondrial endometrial stability decreased, liver damage increased
