目的:研究胃癌及正常胃粘膜c-myc基因外显子ⅢCCGG位点的甲基化状态及其mRNA和蛋白产物p67的表达。方法:应用Southemblot和原位杂交观察14例胃癌和正常胃粘膜c-myc基因DNA甲基化与mRNA表达及SABC法检测胃癌45例、胃溃疡14例、不典型增生10例p67表达。结果:胃癌c-myc基因外显子ⅢCCGG位点比其相应正常胃粘膜呈高度去甲基化,mRNA表达升高(P<0.001),DNA去甲基化与mRNA表达存在相关性(P<0.025)。p67在胃癌、不典型增生、胃溃疡的表达率分别为84.44%(38/45)、100%(10/10)、21.43%(3/14),前两者与后者之间有显著性差异(P<0.001)。结论:c-myc基因外显子ⅢCCGG位点去甲基化及其表达产物可能与胃癌的发生发展有关。
OBJECTIVE: To investigate the methylation status of the exon III CCGG in c-myc gene and expression of mRNA and protein product p67 in gastric cancer and normal gastric mucosa. METHODS: DNA methylation and mRNA expression of c-myc gene were detected in 14 cases of gastric cancer and normal gastric mucosa by Southern blot and in situ hybridization. 45 cases of gastric cancer, 14 cases of gastric ulcer and 10 cases of atypical hyperplasia were detected by SABC method. RESULTS: The c-myc exon III CCGG of gastric cancer was highly demethylated and its mRNA expression was significantly higher than that of the corresponding normal gastric mucosa (P<0.001). There was a correlation between DNA demethylation and mRNA expression. P<0.025). The expression rates of p67 in gastric cancer, atypical hyperplasia, and gastric ulcer were 84.44% (38/45), 100% (10/10), and 21.43% (3/14), respectively. There was a significant difference (P<0.001). Conclusion: The demethylation of c-myc gene exon IIICCGG and its expression products may be related to the occurrence and development of gastric cancer.
