目的观察辛伐他汀在模拟微重力环境下对成骨细胞增殖和凋亡的影响。方法利用沿水平轴连续回转(30 r/min)细胞培养系统模拟微重力环境,使用MTT比色法观察小鼠成骨样细胞MC3T3-E1的增殖情况;细胞凋亡是根据4,6-二氨基-2-苯基吲哚所染细胞核形态的改变来辨认。结果模拟微重力环境可以降低MC3T3-E1增殖功能,辛伐他汀在模拟微重力环境可以增强MC3T3-E1增殖功能,但对细胞的凋亡并无显著影响。结论在模拟微重力环境下辛伐他汀对成骨细胞增殖功能具有保护作用,为辛伐他汀治疗微重力环境骨丢失提供了理论和实验证据。
Objective To observe the effect of simvastatin on proliferation and apoptosis of osteoblasts under simulated microgravity. Methods The microgravity environment was simulated by continuous rotation (30 r / min) cell culture system along the horizontal axis. The proliferation of mouse osteoblast-like cells MC3T3-E1 was observed by MTT assay. The apoptosis was determined by 4,6-bis Amino-2-phenylindole staining of the nuclear morphology to identify changes. Results Simulated microgravity could reduce the proliferation of MC3T3-E1 cells. Simvastatin could enhance the proliferation of MC3T3-E1 cells in simulated microgravity environment but had no significant effect on cell apoptosis. Conclusion Simvastatin has a protective effect on proliferation of osteoblasts under simulated microgravity, which provides theoretical and experimental evidence for simvastatin treatment of bone loss in microgravity environment.
