目的利用响应面法对桑黄菌液体发酵培养基进行优化,提高胞外多糖产量。方法应用Plackett-Burman设计和响应面法,对桑黄菌液体发酵培养基成分及其含量进行优化。用Design-Expert软件对实验数据进行多元回归分析,并建立3种主要因素(蛋白胨、酵母粉和磷酸二氢钾)与多糖产量之间的函数关系。用最终优化的配方进行3次验证实验。结果通过快速“登高法”对3个显著因素进行寻优,培养基中3因素最佳浓度为:蛋白胨14.72 g/L、酵母粉11.04 g/L、磷酸二氢钾1.472 g/L,胞外多糖产量最高可达7.04 g/L。3次验证实验所测得的多糖产量分别为6.92、6.86和7.04 g/L,平均值为6.94 g/L,与预测结果基本一致。结论优化的桑黄菌发酵培养基可显著提高胞外多糖产量。
Objective To optimize the liquid fermentation medium of Phellinus igniarius by response surface methodology to improve the yield of extracellular polysaccharide. Methods Plackett-Burman design and response surface methodology were used to optimize the composition and content of liquid fermentation medium of Phellinus igniarius. Multiple regression analysis of experimental data was performed using Design-Expert software and a functional relationship between the three major factors (peptone, yeast extract and potassium dihydrogen phosphate) and polysaccharide production was established. Three validation experiments with the final optimized formula. Results The three factors were optimized by fast “ascension” method. The optimal concentration of three factors in the medium was: peptone 14.72 g / L, yeast powder 11.04 g / L, potassium dihydrogen phosphate 1.472 g / L, Extracellular polysaccharide production up to 7.04 g / L. The yield of polysaccharides measured by three validation experiments were 6.92, 6.86 and 7.04 g / L, respectively, with an average value of 6.94 g / L, which was in good agreement with the predicted results. Conclusion The optimized fermentation medium of Phellinus igniarius can significantly increase the yield of extracellular polysaccharide.
