目的研究情绪应激大鼠颞下颌关节(TMJ)诱生型一氧化氮合酶(i NOS)在其软骨组织内的表达和作用。方法选择Sprague-Dawley雄性大鼠40只,随机选取10只作为正常组,一直定时喂水,余30只采用空瓶饮水建立大鼠应激模型,在动物应激后的1、3、5周时,分别切取10只大鼠TMJ标本行En Vision两步法免疫组织化学检测i NOS。结果正常组TMJ软骨无或有极少量i NOS表达,应激1周TMJ标本中,已可见i NOS表达;应激3周TMJ标本中表达强烈;应激5周TMJ标本阳性细胞数显著减少且颜色变浅,已呈弱阳性。结论 i NOS在颞下颌关节紊乱发生发展过程中起着重要的作用,抑制i NOS的生成有可能会阻断颞下颌关节紊乱的发生和发展。
Objective To study the expression of iNOS induced by temporomandibular joint (TMJ) in the chondrocytes of emotionally stressed rats. Methods Forty male Sprague-Dawley rats were randomly selected. Ten rabbits were randomly selected as the normal group and fed with water regularly. Thirty rabbits were anesthetized with empty bottle to establish stress model. , 10 rat TMJ specimens were cut out for En Vision two-step immunohistochemical detection of i NOS. Results There was no or very low expression of i NOS in TMJ cartilage in normal group. INOS expression was found in TMJ specimens of 1 week of stress; expression in TMJ specimens at 3 weeks after stress was significantly reduced; and number of TMJ specimens at 5 weeks of stress decreased significantly Light color, has been weak positive. Conclusion i NOS plays an important role in the development of temporomandibular disorder. Inhibiting the production of iNOS may block the occurrence and development of temporomandibular disorder.
