肺部给药是一种简单、直接、有效、无侵害性的给药方式,肺部药物递送可以达到局部及全身的治疗效果。纳米给药系统具有靶向及缓释特性,在提高治疗指数、降低不良反应方面具有显著优势。通过将纳米粒溶液或纳米粒和辅料的混合液进行喷雾干燥,可制得中空性纳米聚集颗粒。中空性纳米聚集颗粒具有较大几何直径和较小的密度,能够提高粒子的空气动力学直径,得到良好的肺部沉积率,是一种潜在的肺部纳米给药系统新剂型。本文从中空性纳米聚集颗粒的形成原理、影响因素、制备工艺等方面阐述了该新型制剂的关键技术,并对其物理化学性质、粉体动力学性质以及用于肺部给药的评价指标进行了总结,为该种新剂型应用于肺部给药的干粉吸入剂提供了制剂学参考。
Pulmonary delivery is a simple, direct, effective, noninvasive method of delivery. Pulmonary drug delivery can achieve both local and systemic therapeutic effects. Nano-drug delivery system with targeted and sustained release characteristics, in improving the therapeutic index, reduce the adverse reaction has a significant advantage. Hollow nano-aggregated particles can be prepared by spray-drying a solution of the nano-particles or a mixture of the nano-particles and the auxiliary materials. Hollow nano-aggregated particles with larger geometric diameter and smaller density, can improve the particle aerodynamic diameter, get a good lung deposition rate, is a potential lung nano-drug delivery system new dosage form. In this paper, the formation of hollow nano-particles aggregates principles, influencing factors, preparation techniques and other aspects of the new formulation of the key technologies, and its physical and chemical properties, powder dynamics and pulmonary delivery for the evaluation of indicators In conclusion, a formulation reference is provided for dry powder inhalers for pulmonary administration of this new dosage form.
