目的探讨红色闪烁光诱导豚鼠近视眼巩膜细胞中结缔组织生长因子(CTGF)的表达及临床意义。方法将60只豚鼠随机分为3组:Ⅰ组红色闪烁光照射(完全密闭的纸箱);Ⅱ组正常自然光照射(四周密闭而上方开放的纸箱);Ⅲ组正常自然光照射(视野开阔的笼中)。实验前和实验8周后,睫状肌麻痹下验光、A超动态观察豚鼠眼屈光度和眼轴长度变化,用免疫组织化学染色法和Western-blot法检测豚鼠后极部巩膜CTGF的表达。结果Ⅰ屈光度明显下降,且伴有眼轴延长。Ⅰ、Ⅱ、Ⅲ组巩膜中均有CTGF的表达,Ⅰ组巩膜CTGF的表达量明显比Ⅱ、Ⅲ组高。结论红色闪烁光诱导豚鼠近视眼后极部巩膜中CTGF表达增强,CTGF可能在近视发展中发挥重要作用。
Objective To investigate the expression of connective tissue growth factor (CTGF) in the scleral cells of guinea pig myopic eyes induced by red light and its clinical significance. Methods Sixty guinea pigs were randomly divided into three groups: group I was irradiated with red flashing light (fully enclosed carton); group II was exposed to natural light (closed and the cartons were open at the top); group III was exposed to natural light ). Eight weeks before and after the experiment, the ciliary muscle paralysis under the light, A dynamic observation of guinea pig eyes refraction and axial length changes, immunohistochemical staining and Western-blot assay guinea pig posterior scleral CTGF expression. Results Ⅰ diopter decreased significantly, and accompanied by axial extension. The expression of CTGF in the sclera of group Ⅰ, Ⅱ and Ⅲ was significantly higher than that in group Ⅱ and Ⅲ. Conclusion The red scintigraphy induced CTGF expression in posterior sclera of guinea pig myopic eyes. CTGF may play an important role in the development of myopia.
