目的:观察尿毒清对单侧输尿管梗阻(unilateral ureteral obstruction,UUO)大鼠肾脏转化生长因子β激活激酶(TGFβ-activated kinase 1,TAK1)及纤维黏连蛋白(fibronectin,FN)表达的影响。方法:采用单侧输尿管结扎术制备肾间质纤维化大鼠模型,将72只Sprague-Wistar(SD)大鼠随机分为3组:假手术组、UUO模型组及尿毒清治疗组,每组24只,治疗组于术前1 d给予尿毒清2 g.kg-1.d-1,灌胃(ig),假手术组与UUO模型组每天ig等量生理盐水。各组分别于术后7,14,21 d处死8只大鼠,采用HE及Masson染色观察梗阻侧肾脏病理变化,原位杂交及RT-PCR检测肾组织TAK1 mRNA的表达,Western blot法检测TAK1及FN蛋白的表达。结果:与假手术组比较,模型组及治疗组TAK1,FN表达均增高(P<0.01),而且模型组显著高于治疗组(P<0.05)。结论:尿毒清可能通过下调TAK1及FN的表达,从而减轻肾间质纤维化。
Objective: To observe the effect of Niaoduqing on the expression of TGFβ-activated kinase 1 (TAK1) and fibronectin (FN) in rats with unilateral ureteral obstruction (UUO). Methods: A rat model of renal interstitial fibrosis was established by unilateral ureteral ligation. Seventy-two Sprague-Wistar rats were randomly divided into three groups: sham-operated group, UUO model group and nimodipine-treated group. Twenty-four rabbits in the treatment group were treated with 2 g.kg-1.d-1 of nifedipine one day before operation, and the rats in the treatment group and the UUO model group were given the same amount of normal saline every day. The rats in each group were sacrificed on the 7th, 14th and 21st day after the operation, respectively. Eight rats were sacrificed and pathological changes of obstructive kidneys were observed by HE and Masson staining. The expression of TAK1 mRNA in renal tissues was detected by in situ hybridization and RT-PCR. And FN protein expression. Results: Compared with the sham operation group, the expressions of TAK1 and FN in model group and treatment group were significantly increased (P <0.01), and the model group was significantly higher than the treatment group (P <0.05). Conclusion: Niaoduqing may reduce renal interstitial fibrosis by down-regulating the expression of TAK1 and FN.
