目的:探讨舒心通脉片改善异丙肾上腺素介导的心肌缺血的作用机制与其抑制含Nox4的NADPH氧化酶介导的氧化应激。方法:研究舒心通脉片对异丙肾上腺素诱导的心肌缺血大鼠心电图和左心室Nox4表达水平的影响,以及大鼠给药后血清中乳酸脱氢酶(lactate dehydrogenase,LDH)、磷酸肌酸激酶(creatine phosphokinase,CK)、超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)浓度的变化。结果:舒心通脉片可抑制异丙肾上腺素介导的心肌缺血大鼠中ST段位移值的增加,降低血清中LDH和CK水平,升高SOD活性,降低左心室中Nox4的表达水平。结论:舒心通脉片可改善异丙肾上腺素诱导的心肌缺血,其机制可能与降低Nox4介导的活性氧的产生和升高SOD活性有关。
Objective: To investigate the mechanism of Shuxintongmai tablet in improving isoproterenol-mediated myocardial ischemia and its inhibition of NADPH oxidase-mediated oxidative stress. Methods: The effects of Shuxintongmai tablet on ECG and left ventricular Nox4 expression in isoproterenol-induced myocardial ischemia rats were studied. The levels of lactate dehydrogenase (LDH) Changes in the levels of creatine phosphokinase (CK), superoxide dismutase (SOD) and malondialdehyde (MDA) were observed. Results: Shuxin Tongmai tablet could inhibit the increase of ST segment displacement in isoproterenol-induced myocardial ischemia rats, decrease the level of LDH and CK in serum, increase the activity of SOD and decrease the expression of Nox4 in the left ventricle. Conclusion: Shuxintongmai tablet can improve isoproterenol-induced myocardial ischemia, and its mechanism may be related to reducing Nox4-mediated reactive oxygen species production and increasing SOD activity.
