本实验用荧光染色法和流式细胞技术 ,研究苯对小鼠外周血淋巴细胞凋亡的诱导作用。发现苯体外诱导小鼠外周血淋巴细胞产生典型的凋亡形态学改变 ,用不同浓度的苯诱导淋巴细胞 3小时 ,随苯浓度的不同 ,细胞凋亡率也有相应变化 ,呈现出剂量效应关系 ,用浓度为 10 mmol/ L 的苯诱导淋巴细胞 ,随诱导时间不同细胞凋亡率也有相应变化。流式细胞技术检测结果与荧光染色法结果基本吻合。提示 :苯对体外培养的小鼠外周血淋巴细胞具有凋亡诱导作用。
In this study, fluorescence staining and flow cytometry were used to study the induction of benzene on peripheral blood lymphocyte apoptosis in mice. The results showed that benzene induced morphological changes of typical peripheral blood lymphocytes in vitro. With different concentrations of benzene induced lymphocytes for 3 hours, with the different concentrations of benzene, the apoptosis rate also showed a dose-response relationship, With the concentration of 10 mmol / L of benzene induced lymphocytes, with the induction of apoptosis rate of different cells also have a corresponding change. Flow cytometry results and fluorescence staining results basically consistent. Tip: benzene on mouse peripheral blood lymphocytes with apoptosis-inducing effect.
