目的:观察小泛素化相关修饰蛋白质类(SUMO)及SUMO化修饰蛋白在恶性胶质瘤组织中的表达,阐明SUMO化与恶性胶质瘤发生的关系。方法:人脑正常组织和恶性胶质瘤组织样品分别作为正常对照组和恶性胶质瘤组,采用蛋白免疫印迹法检测SUMO1、SUMO2、SUMO活化酶(Aos1和Uba2)、SUMO结合酶(UBC9)和SUMO连接酶(PIAS1、PIAS2、PIAS3和Pc2)蛋白的表达水平。结果:与正常对照组比较,恶性胶质瘤组织中SUMO1、SUMO2、Aos1、UBC9和PIAS3蛋白表达水平明显升高(P<0.01),Uba2、PIAS1和PIAS2蛋白表达水平明显降低(P<0.01)。结论:SUMO及SUMO化修饰蛋白可能对恶性胶质瘤的发生起重要的促进作用。
Objective: To observe the expression of SUMO and SUMO-modified proteins in malignant glioma and to elucidate the relationship between SUMO and malignant glioma. Methods: Normal human brain tissue and malignant glioma tissue samples were used as normal control group and malignant glioma group. The expressions of SUMO1, SUMO2, SUMO-activated enzyme (Aos1 and Uba2), SUMO-binding enzyme (UBC9) And SUMO ligase (PIAS1, PIAS2, PIAS3 and Pc2) protein expression levels. Results: Compared with the normal control group, the expression of SUMO1, SUMO2, Aos1, UBC9 and PIAS3 in malignant glioma tissues were significantly increased (P <0.01), and the expressions of Uba2, PIAS1 and PIAS2 protein were significantly decreased . Conclusion: SUMO and SUMO-modified protein may play an important role in the progression of malignant glioma.
