目的通过研究a-B crystallin在不同恶性程度乳腺癌细胞中的表达程度,阐明a-B crystallin在乳腺癌临床应用中的意义。方法①采用逆转录聚合酶链(RT-PCR)技术检测不同恶性程度乳腺癌细胞株MCF-7(高)、MDA-MB-453(中)、MDA-MB-231(低)中a-B crystallin基因的表达。②采用Western blot检测在三种乳腺癌细胞株中a-B crystallin蛋白的表达。结果①a-B crystallin基因的表达:在三种乳腺癌细胞株中a-B crystallin随恶性程度的增高,其基因表达量明显降低,表达比例为4∶2∶1(P<0.01)。②a-B crystallin蛋白表达:在三种乳腺癌细胞株中a-Bcrystallin随恶性程度的增高其蛋白表达量明显降低,表达比例为3.5∶1.9∶1(P<0.01)。结论常规检测乳腺癌中a-B crystallin基因和蛋白表达水平,并与其他指标结合,有助于判断乳腺癌细胞的恶性程度及转移性。
Objective To study the expression of a-B crystallin in breast cancer with different degrees of malignancy by studying the expression of a-B crystallin in clinical application of breast cancer. Methods ① Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the gene aB crystallin in MCF-7, MDA-MB-453 and MDA-MB- expression. Western blot was used to detect the expression of a-B crystallin protein in three breast cancer cell lines. Results ① The expression of a-B crystallin gene: The gene expression of a-B crystallin decreased with the increase of the malignant degree in the three breast cancer cell lines, the expression ratio was 4: 2:1 (P <0.01). ② a-B crystallin protein expression: a-Bcrystallin in three breast cancer cell lines with a significant increase in the degree of its protein expression was significantly reduced, the expression ratio of 3.5: 1.9: 1 (P <0.01). Conclusion Conventional detection of a-B crystallin gene and protein expression in breast cancer, combined with other indicators, helps to determine the degree of malignancy and metastatic breast cancer cells.
