目的:研究葡萄糖转运体(Glut1)在大肠癌细胞系及临床样本中表达水平,并探讨其与大肠癌发生发展的相关性。方法采用Western blotting法分别检测大肠癌细胞系及大肠癌组织中Glut1蛋白表达水平;采用免疫组化方法检测Glut1蛋白表达水平;采用葡萄糖检测试剂盒检测大肠癌细胞葡萄糖的消耗。结果 Western blot结果显示:与癌旁组织相比, Glut1的表达水平明显升高(P<0.05);与正常肠粘膜细胞相比,大肠癌细胞系中Glut1的表达水平明显升高。免疫组化结果显示:与癌旁组织相比, Glut1的表达水平明显升高(P<0.05),而且其表达水平与大肠癌的病理分期密切相关(P<0.05)。葡萄糖检测结果显示:与正常大肠粘膜细胞相比,大肠癌细胞培养液中葡萄糖的含量明显减少。结论 Glut1的表达水平与大肠癌的发生发展成正相关。“,”Objective To study the expression of glucose transporter 1 ( Glut 1 ) in colon cancer cell lines and clinical samples and to explore the correlation between colon cancer progression and the expression. Methods The Glut1 protein expression in colon cancer tissues and cell lines was determined with Western blotting and immunohistochemisty respectively. The consumption of glucose in colon cancer cells was examined with Glucose ( GO) Assay Kit. Results Western blotting results showed that the expression of Glut1 in colon cancer cell lines significantly increased compared with that in tumor-adjacent tissues and in normal intestinal mucosa cells (P<0. 05). As immunohistochemisty results showed, the expression of Glut1 in cancer tissues increased remarkably compared with that in tumor-adjacent tissues (P<0. 05), and the expression was closely related to the pathological stage of colon cancer (P<0. 05). The results of Glucose (GO) Assay Kit suggested that the content of glucose in the culture media of colon cancer cell lines de-creased significantly compared with that in normal intestinal mucosa cells. Conclusion Glut1 expression is positively related to colon cancer progression.
