目的:分离、鉴定肺癌干细胞,并探讨肺岩宁方对肺癌干细胞Wnt信号通路的干预作用。方法:分离、鉴定肺癌干细胞;将分离出的SP+肺癌干细胞分为对照组(空白血清+生理盐水)、单纯化疗(空白血清+DDP组)、肺岩宁方组(肺岩宁方含药血清+生理盐水)、综合治疗组(肺岩宁方含药血清+DDP),予体外药物干预后检测β-catenin蛋白表达及SP+肺癌干细胞凋亡情况。结果:分离的SP+、CD24+IGF-1R+、CD133+细胞具有肺癌干细胞特性,分离的β-catenin蛋白完整;综合治疗组诱导SP+干细胞凋亡优于其他组,各组凋亡率分别为综合治疗组79.2%、肺岩宁方组21.3%、DDP组33.7%、对照组0.3%。结论:本课题组已掌握分离、鉴定肺癌干细胞的技术,中药肺岩宁方联合化疗具有一定的诱导肺癌干细胞凋亡的作用。
OBJECTIVE: To isolate and identify lung cancer stem cells and explore the effect of Feiyanning Fang on Wnt signaling pathway in lung cancer stem cells. Methods: The lung cancer stem cells were isolated and identified. The SP + lung cancer stem cells isolated were divided into control group (blank serum + saline), chemotherapy alone (blank serum + DDP group), Feiyanningfang group + Saline) and the combination therapy group (Feiyanning Decoction + DDP) were used to detect the expression of β-catenin protein and the apoptosis of SP + lung cancer stem cells in vitro. Results: The isolated SP +, CD24 + IGF-1R + and CD133 + cells had the characteristics of lung cancer stem cells, and the isolated β-catenin protein was intact. The apoptosis of SP + stem cells in combination therapy group was better than other groups. 79.2% in lung Yanning group, 21.3% in lung Yanning group, 33.7% in DDP group and 0.3% in control group. Conclusion: Our group has mastered the technology of isolation and identification of lung cancer stem cells. The combination of Chinese herbal medicine Feiyanning prescription and chemotherapy can induce the apoptosis of lung cancer stem cells.
