[目的]研究顺铂(DDP)与5-氟尿嘧啶(5-FU)在合并用药前后对体外培养的人胃癌细胞的影响。[方法]采用MTT法观察体外培养的人胃癌细胞系SGC-7901在DDP与5-FU合并用药前后细胞存活率的差异,并通过流式细胞仪检测细胞周期变化,透射电镜观察细胞超微结构变化。[结果]DDP与5-FU合用与单独应用化疗药相比细胞存活率明显下降,其增效倍数为单独用药的2.73~4.48倍(P<0.01),细胞生长停止在S期,S期数分数为0.51,有效抑制了细胞有丝分裂及DNA合成,细胞超微结构显示:联合用药后,细胞核固缩、空泡形成、微绒毛减少、线粒体及内质网肿胀。[结论]结果表明顺铂与小剂量化疗药合用,可获得强于化疗药单独用药的疗效,5-氟尿嘧啶增加了化疗药物对肿瘤细胞的毒性,增加了细胞对药物的敏感性,降低了药物的副作用。
[Objective] To study the effect of DDP and 5-FU on human gastric cancer cells cultured in vitro before and after combination therapy. [Methods] The cell viability of human gastric cancer cell line SGC-7901 cultured in vitro was observed by MTT assay before and after the combination of DDP and 5-FU. [Conclusion] The results show that cisplatin combined with low-dose chemotherapy can be obtained stronger than chemotherapy alone, 5-fluorouracil increases the toxicity of chemotherapeutic drugs on tumor cells, increasing the sensitivity of cells to drugs, reducing the drug Side effects.
