【目的】建立蜂蜜中氨基酸成分的高效液相(HPLC)指纹图谱分析方法。【方法】采用732阳离子交换树脂富集蜂蜜中的氨基酸,而后采用异硫氰酸苯酯柱前衍生法,以脯氨酸为参照峰。色谱柱为Waters Symmetry C18色谱柱(4.6 mm×250mm,5μm);流动相:A为乙腈∶水(体积比为4∶1),B为30 mmol/L醋酸钠溶液∶乙腈(体积比为355∶15),用冰乙酸调节p H=6.5,梯度洗脱;检测波长:254 nm;流速:1.0 m L/min;进样量5μL。【结果】15批蜂蜜样品氨基酸指纹图谱共标出16个共有峰,15批样品的相似度在0.910~0.996之间。【结论】该指纹图谱检测方法简便可行、重现性好、特征性强,可为蜂蜜质量控制提供一定参考。
【Objective】 HPLC method for the determination of amino acids in honey was established. 【Method】 The 732 cation exchange resin was used to enrich the amino acids in honey, then the phenylisothiocyanate pre-column derivatization method was used, and proline was used as the reference peak. The chromatographic column was a Waters Symmetry C18 column (4.6 mm × 250 mm, 5 μm); the mobile phase consisted of acetonitrile: water (volume ratio 4: 1), B 30 mmol / L sodium acetate solution : 15), adjusted to pH 6.5 with glacial acetic acid, gradient elution; detection wavelength: 254 nm; flow rate: 1.0 mL / min; injection volume 5 μL. 【Result】 A total of 16 common peaks were identified by amino acid fingerprint in 15 batches of honey samples. The similarity of 15 batches of samples was between 0.910 and 0.996. 【Conclusion】 The fingerprint detection method is simple and feasible, reproducible, strong characteristic, and can provide some reference for the quality control of honey.
