目的 :探讨薯蓣皂苷元(Dio)对野百合碱(MCT)诱导的肺动脉高压大鼠肺血管重构的影响。方法 :将30只雄性SD大鼠随机分为正常对照组、MCT组和Dio组,每组10只。MCT组和Dio组一次性腹腔注射野百合碱60mg/kg,建立大鼠肺动脉高压模型。Dio组每天给予Dio 80mg/kg灌胃,对照组和MCT组给予相同量溶剂灌胃。待药物干预满4周后,分别测量各组干预后平均肺动脉压力(mPAP)、右心室肥大指数(RVHI)、肺小动脉管壁厚度占管径的百分比(WT%)和管壁面积占血管总面积的百分比(WA%);采用免疫组化检测肺组织核因子-κB(NF-κB)的表达。结果 :MCT组的m PAP、RVHI、WT%、WA%及NF-κB的表达水平均显著高于对照组和Dio组;经薯蓣皂苷元干预后,m PAP、RVHI、WT%、WA%及NF-κB的表达水平均明显降低。结论 :薯蓣皂苷元能够缓解MCT诱导的肺动脉高压大鼠肺血管重构,其机制可能与抑制NF-κB的表达有关。
Objective: To investigate the effect of diosgenin (Dio) on pulmonary vascular remodeling induced by monocrotaline (MCT) in rats with pulmonary hypertension. Methods: Thirty male Sprague-Dawley rats were randomly divided into normal control group, MCT group and Dio group, with 10 rats in each group. Rats in MCT group and Dio group were given intraperitoneal injection of monocrotaline (60mg / kg) to establish a model of pulmonary hypertension in rats. The Dio group was orally administered with 80 mg / kg of Dio daily, while the control group and the MCT group were administered with the same amount of solvent. Four weeks after drug intervention, mean pulmonary arterial pressure (mPAP), right ventricular hypertrophy index (RVHI), percentage of pulmonary arteriole wall thickness to tube diameter (WT%), and wall area of blood vessels The percentage of the total area (WA%). The expression of nuclear factor-κB (NF-κB) was detected by immunohistochemistry. Results: The levels of m PAP, RVHI, WT%, WA% and NF-κB in MCT group were significantly higher than those in control group and Dio group. The m PAP, RVHI, WT%, WA% NF-κB expression levels were significantly lower. Conclusion: Diosgenin can relieve pulmonary vascular remodeling induced by MCT in rats with pulmonary hypertension, which may be related to the inhibition of NF-κB expression.
