目的探讨雌激素改善大鼠记忆与BDNF表达的关系。方法停止繁殖的15月龄雌性大鼠,体质量350~400g,随机分为治疗组和对照组,在颈部皮下分别注射相同剂量的苯甲酸雌二醇和生理盐水。采用Morris水迷宫检测大鼠记忆,大鼠海马BDNF、TrkB及mRNA表达采用RT-PCR检测,BDNF、TrkB及proBDNF蛋白表达采用Western-blot检测。结果治疗组大鼠潜伏期缩短,海马BDNF、TrkB、proBDNF表达上调(P<0.01)。结论雌激素改善大鼠记忆,可能与BDNF及相关蛋白表达上调有关。
Objective To explore the relationship between estrogen and memory of rats and the expression of BDNF in rats. Methods Female 15-month-old female rats whose body weight was 350-400g were randomly divided into treatment group and control group. The same dose of estradiol benzoate and normal saline were injected subcutaneously into the neck respectively. The Morris water maze test was used to detect the memory of rat hippocampal BDNF, TrkB and mRNA expression by RT-PCR detection, BDNF, TrkB and proBDNF protein expression was detected by Western-blot. Results The latency of treatment group was shortened and the expression of BDNF, TrkB and proBDNF in hippocampus was increased (P <0.01). Conclusion Estrogen may improve the memory of rats, which may be related to the up-regulation of BDNF and related proteins.
