目的应用巢式PCR方法对低模板量DNA的流脑样本进行MLST分型检测。方法根据脑膜炎奈瑟菌MLST分型检测所选择的管家基因序列选择巢式PCR所需要的引物,对低模板量DNA的流脑样本进行MLST分型检测。结果对6份血清、脑脊液标本进行MLST分型检测,其中3份标本检测成功。结论巢式PCR方法提高了低模板量DNA的流脑样本的MLST分型检测的敏感性。
Objective To apply nested PCR method to detect MLST typing of low-template DNA in meningitis. Methods According to the selected housekeeping gene sequence of Neisseria meningitidis MLST genotyping, primers required for nested PCR were selected and MLST typing was performed on meningococcal meningitis samples with low template DNA. Results Six serum and cerebrospinal fluid samples were tested by MLST, of which three were successfully detected. Conclusion The nested PCR method increases the sensitivity of MLST typing in meningococcal samples with low template DNA.
