目的:探讨黄芩素对MG63细胞侵袭浸润能力的影响,并对其机制进行初步探讨。方法:MG63细胞分为对照组和黄芩素组,于培养液中加入黄芩素使其终浓度分别为50、100、200μmol/L。分别采用real time PCR、Elisa方法检测肿瘤组织中MMP-2、整合素α2 mRNA和蛋白表达量;采用Boyden小室法观测黄芩素对MG63细胞侵袭能力的影响。结果:黄芩素50μmol/L能明显降低MG63细胞中MMP-2、整合素α2 mRNA和蛋白含量;黄芩素100、200μmol/L能显著降低MMP-2、整合素α2 mRNA和蛋白含量;黄芩素50、100、200μmol/L能显著降低MG63细胞的穿膜细胞数。结论:黄芩素能降低MG63细胞的侵袭转移能力,其机制可能与降低MMP-2、整合素α2表达有关。
Objective: To investigate the effect of baicalein on invasion and infiltration of MG63 cells, and to explore its mechanism. Methods: MG63 cells were divided into control group and baicalein group. Baicalein was added to the culture medium to make the final concentrations of 50, 100, 200 μmol/L. Real-time PCR and Elisa methods were used to detect the expression of MMP-2 and integrin α2 mRNA and protein in tumor tissue. The Boyden chamber method was used to observe the effect of baicalein on the invasive ability of MG63 cells. Results: Baicalein 50μmol/L can significantly reduce the content of MMP-2 and integrin α2 mRNA and protein in MG63 cells. Baicalein 100 and 200μmol/L can significantly reduce the content of MMP-2 and integrin α2 mRNA and protein; Baicalein 50 , 100, 200μmol/L can significantly reduce the number of MG63 cell transmembrane cells. Conclusion: Baicalein can reduce the invasion and metastasis of MG63 cells, and its mechanism may be related to the decrease of MMP-2 and integrin α2 expression.
