HCV(hepatitis C virus)是引起丙型肝炎的病原体,丙型肝炎病程长者可达30年以上,最终可发展成肝硬化及肝细胞肝癌。早期确诊HCV感染和及时有效地治疗,是目前世界上肝脏疾病研究的重要课题。本研究建立了灵敏性强、特异性高的PCR方法,逆转录扩增HCV严密保守区5’-非编码区基因,该靶区位置于-276-21间,得到256bp片断。灵敏性可达10~1~10~2HCV拷贝/ml。检测120例临床诊断为非甲非乙型肝炎患者,其PCR结果与第二代ELISA检测血清抗体相比较,ELISA阳性中95.4%PCR检测阳性,而ELISA阴性者中8.9%PCR检测阳性。分析了63株临
Hepatitis C virus (HCV) is the causative agent of hepatitis C, and the course of hepatitis C is up to 30 years old, eventually leading to cirrhosis and hepatocellular carcinoma. Early diagnosis of HCV infection and timely and effective treatment, is the world’s major issue of liver disease research. In this study, a highly sensitive and specific PCR method was established to reverse-amplify the 5 ’noncoding region of HCV tight conserved region. The target region was located between -276-21 and a 256 bp fragment was obtained. Sensitivity up to 10 ~ 1 ~ 2 ~ 2HCV copies / ml.
