目的:检测NOMO1基因在SD大鼠胚胎心脏发育过程中mRNA表达变化及定位,并探讨其在胚胎心脏发育的意义。方法:24只孕鼠随机分为6组,每组4只,分别于妊娠12 d(D12)、15 d(D15)、16 d(D16)、17 d(D17)、19 d(D19)、21 d(D21)时取出胎鼠,取其心脏,采用实时定量PCR(real-time PCR)技术检测心脏组织中NOMO1基因mRNA的表达丰度,并应用荧光原位杂交(FISH)的方法对其进行定位分析。结果:在妊娠D12、D15、D16、D17、D19、D21的胚胎心脏中NOMO1基因mRNA均有表达,且呈由弱到强再减弱趋势,表达高峰出现在D16;该基因主要表达于心外膜中,而在心肌层、心内膜、心瓣膜等处未见表达。结论:NOMO1基因的表达随大鼠胚胎的心脏发育呈动态表达,可能与心脏发育相关。
OBJECTIVE: To detect the mRNA expression and location of NOMO1 gene during embryonic heart development in SD rats and to explore its significance in embryonic heart development. Methods: Twenty-four pregnant rats were randomly divided into 6 groups with 4 rats in each group. The pregnant rats were randomly divided into 6 groups at D12, D15, D16, D17 and D19, The fetus was taken out on day 21 (D21), and the heart was taken out. The mRNA expression of NOMO1 in heart tissue was detected by real-time PCR, and its expression was detected by fluorescence in situ hybridization (FISH) Positioning analysis. Results: NOMO1 mRNA was expressed in embryonic heart of D12, D15, D16, D17, D19 and D21, and then decreased from weak to strong. The peak of expression was at D16. The gene was mainly expressed on the epicardium In the myocardium, endocardium, heart valves and other places no expression. CONCLUSION: The expression of NOMO1 gene is dynamically expressed in the heart of rat embryos, which may be related to cardiac development.
