目的:检测桥本甲状腺炎患者外周血中趋化因子CCL20表达水平,探讨CCL20水平与TPOAb和TgAb的相关性。方法:TRIzol提取32例桥本甲状腺炎患者(甲状腺炎组)及22例健康人(对照组)外周血中淋巴细胞总RNA,real-time PCR检测CCL20 mRNA表达水平;用酶联免疫吸附法(ELISA)检测外周血中CCL20蛋白表达水平,统计学方法分析CCL20表达水平与TPOAb和TgAb的相关性。结果:甲状腺炎组外周血中CCL20表达高于对照组(P<0.05);CCL20表达水平与TPOAb和TgAb呈正相关(P<0.05)。结论:桥本甲状腺炎患者外周血中CCL20表达增高,可能参与桥本甲状腺炎的发病。
Objective: To detect the expression of chemokine CCL20 in peripheral blood of patients with Hashimoto’s thyroiditis and to explore the correlation between CCL20 level and TPOAb and TgAb. Methods: The total RNA of peripheral blood lymphocytes from 32 Hashimoto’s thyroiditis patients (thyroiditis group) and 22 healthy volunteers (control group) were extracted by TRIzol. The expression of CCL20 mRNA was detected by real-time PCR. The expression of CCL20 mRNA was detected by enzyme-linked immunosorbent assay ELISA) was used to detect the expression of CCL20 protein in peripheral blood. The correlation between CCL20 expression level and TPOAb and TgAb was analyzed statistically. Results: The expression of CCL20 in thyroiditis group was higher than that in control group (P <0.05). The expression of CCL20 was positively correlated with TPOAb and TgAb (P <0.05). Conclusion: The expression of CCL20 in peripheral blood of patients with Hashimoto’s thyroiditis is increased, which may be involved in the pathogenesis of Hashimoto’s thyroiditis.
