目的建立一种用声光可调(AOTF)-近红外光谱技术快速测定安宫牛黄丸中黄芩苷含量的新方法。方法以HPLC分析值作参比,运用偏最小二乘法(PLS1)法建立黄芩苷NIR光谱信息与含量间的定量校正模型。结果安宫牛黄丸混合药粉模型中,黄芩苷的内部交叉验证均方差(RMSEC)是0.3213,决定系数是0.9897,黄芩苷预测值与真值的相关系数是0.9806。安宫牛黄丸待包丸模型中,黄芩苷的内部交叉验证均方差(RMSEC)是0.0785,决定系数是0.9501,黄芩苷预测值与真值的相关系数是0.9751。结论AOTF-近红外光谱法对安宫牛黄丸生产过程中黄芩苷的含量预测结果较好,可为中药生产过程的在线、无损定量分析提供依据。
Objective To establish a new method for rapid determination of baicalin in Angong Niuhuang Wan by AOTF - NIR spectroscopy. Methods HPLC analysis was used as a reference, and the PLS1 method was used to establish a quantitative calibration model of the information and content of baicalin NIR spectra. Results In the Angong Niuhuang pill mixed powder model, the internal cross validation of baicalin was 0.3213, the determination coefficient was 0.9897, and the correlation coefficient between baicalin prediction and true value was 0.9806. In the Angong Niuhuang Wanqun Pill Pill model, the internal cross-validation mean baicalin (RMSEC) of baicalin was 0.0785, the determination coefficient was 0.9501, and the correlation coefficient between baicalin prediction and true value was 0.9751. Conclusion AOTF-NIRS has a good predictive value for the content of baicalin in the production of Angong Niuhuang Wan, which can provide the basis for the online and non-destructive quantitative analysis of the production process of Chinese traditional medicine.
