目的检测新的长链非编码RNA(LncRNA)T-ALL-R-LncR1在胚胎型横纹肌肉瘤(ERMS)中的表达。方法对BALB/c裸鼠皮下注射RD细胞悬液建立ERMS动物模型;成瘤后肿瘤组织行HE染色和免疫组织化学检测,镜下观察结果。RT-PCR法检测T-ALL-R-LncR1在RD细胞和ERMS动物模型肿瘤组织中的表达。结果接种RD细胞的裸鼠全部出现移植瘤,成瘤率100%。镜下观察HE染色及免疫组织化学切片后确定肿瘤组织病理分型为ERMS。通过RT-PCR检测及测序在RD细胞和ERMS动物模型肿瘤组织中发现T-ALL-R-LncR1。结论成功建立ERMS动物模型。新的LncRNA T-ALL-R-LncR1可能为今后研究ERMS的诊断及治疗提供了新的靶标。
Objective To detect the expression of a novel long-chain non-coding RNA (Tn-LncRNA) T-ALL-R-LncR1 in embryonal rhabdomyosarcoma (ERMS). Methods BALB / c nude mice were injected subcutaneously with RD cell suspension to establish an animal model of ERMS. Tumor tissues after tumor formation were examined by HE staining and immunohistochemistry, and the results were observed microscopically. RT-PCR was used to detect the expression of T-ALL-R-LncR1 in RD cells and ERMS animal models. Results All the nude mice inoculated with RD cells developed xenografts and the tumorigenic rate was 100%. Microscopically, HE staining and immunohistochemical staining were performed to determine the histopathological type of the tumor as ERMS. T-ALL-R-LncR1 was found in RD cells and in ERMS animal model tumor tissues by RT-PCR detection and sequencing. Conclusion The ERMS animal model was successfully established. The new LncRNA T-ALL-R-LncR1 may provide a new target for the future study of the diagnosis and treatment of ERMS.
