目的探讨结肠癌细胞OPN和HIF-1a基因表达水平与结肠癌细胞恶性生活学行为的关系。方法培养人结肠癌细胞,采用荧光定量PCR法检测患者细胞中OPN和HIF-1a的表达水平,并检测细胞增殖、迁移、侵袭功能。结果腺病毒转染后12 h、24 h、36 h、48 h,rAd-OPN组和rAd-HIF-1a组细胞中OPN和HIF-1a基因表达mRNA含量均高于rAd-NC组,rAd-OPN组和rAd-HIF-1a组细胞迁移率均高于rAd-NC组,rAd-OPN和rAd-HIF-1a组的穿膜细胞数目均多于rAd-NC组,以上差异均有统计学意义(P<0.05)。结论 OPN和HIF-1a基因高表达的重组腺病毒能够抑制结肠癌恶性生物学行为。
Objective To investigate the relationship between the expression of OPN and HIF-1a in colon cancer cells and the malignant behavior of colon cancer cells. Methods Human colon cancer cells were cultured. The expression of OPN and HIF-1a in the cells were detected by real-time PCR, and the function of proliferation, migration and invasion were detected. Results The mRNA expression of OPN and HIF-1a mRNA in rAd-OPN group and rAd-HIF-1a group were higher than those in rAd-NC group at 12 h, 24 h, 36 h and 48 h after transfection, The cell migration rates in OPN group and rAd-HIF-1a group were higher than those in rAd-NC group, and the number of transmembrane cells in rAd-OPN and rAd-HIF-1a groups were more than rAd-NC group, the above differences were statistically significant (P <0.05). Conclusion The recombinant adenovirus overexpressing OPN and HIF-1a gene can inhibit the malignant behavior of colon cancer.
