目的观察揉髌手法对兔膝关节骨关节炎模型血清代谢组学的影响,探讨其治疗的可能作用机制。方法 60只新西兰兔分为手法组、对照组、假模组和正常组,各15只,手法组、对照组手术造成骨内高压型动物模型,假模组手术但不损伤相应血管,正常组不做任何处理。手法组每只兔揉髌手法治疗17次,其他兔无特殊处理。造模后10天、12周末耳缘静脉取血,核磁共振波谱仪检测血清样本,软件进行PCA和PLS-DA分析。结果血清检测分析显示手法组与模型组从术后10天到第12周末在PCA得分图上有分离趋势;PLS-DA载荷图提示手法组、模型组和正常组术后第12周末的差异性代谢物有羟嘌呤醇、乙酸盐、甜菜碱、甘氨酸、脯氨酸、肌酐、瓜氨酸、柠檬酸盐、苏氨酸、谷氨酰胺、精氨酸、赖氨酸、丙氨酸等。结论揉髌手法延缓膝关节软骨组织的退变可能与血清代谢物的改变有关。
Objective To observe the effect of kneading patellar approach on serum metabonomics of rabbit knee osteoarthritis model and to explore the possible mechanism of action. Methods Sixty New Zealand white rabbits were divided into three groups: hand group, control group, fake model group and normal group, 15 cases in each group. Manipulation group and control group caused intraosseous high pressure animal model and fake model group operation without damaging corresponding blood vessel, normal group Do not do anything. Each rabbit was kneaded patella manipulation 17 times, no special treatment of other rabbits. Blood samples were taken from the veins of the ears at 10 days after modeling and 12 weeks later. Serum samples were detected by nuclear magnetic resonance spectroscopy and analyzed by PCA and PLS-DA software. Results The results of serum test showed that there was a trend of separation on the PCA score map from the 10 days to the 12th weekend in the manipulation group and the model group. The PLS-DA load map showed the difference between the 12th weekend after surgery, the model group and the normal group Metabolites have oxypurinol, acetate, betaine, glycine, proline, creatinine, citrulline, citrate, threonine, glutamine, arginine, lysine, alanine, etc. . Conclusion Kneading patellar manipulation delaying the degeneration of cartilage in knee joint may be related to the change of serum metabolites.
