目的研究肾康注射液对单侧输尿管梗阻(UUO)大鼠模型肾间质转化生长因子-β1(TGF-β1)和骨形成蛋白-7(BMP-7)表达的影响。方法将SD雌性大鼠54只随机分为正常对照组、UUO模型组、肾康治疗组,肾康治疗组用肾康注射液灌胃,分别在手术后第5,10,15天处死大鼠,在光镜下观察肾组织病理改变,应用免疫组织化学方法检测肾小管间质中TGF-β1和BMP-7表达。结果与正常对照组相比,UUO模型组大鼠肾脏TGF-β1表达明显升高,BMP-7表达明显减少,但在予肾康注射液治疗后能显著下调UUO模型大鼠TGF-β1表达,上调BMP-7表达。结论肾康注射液可减轻肾小管间质纤维化,延缓慢性肾功能衰竭进展。
Objective To study the effect of Shenkang injection on the expression of transforming growth factor-β1 (TGF-β1) and bone morphogenetic protein-7 (BMP-7) in renal interstitial unilateral ureteral obstruction (UUO) rats. Methods 54 SD female rats were randomly divided into normal control group, UUO model group, and Shenkang treatment group. Shenkang treatment group was treated with Shenkang injection. Rats were sacrificed at 5, 10, and 15 days after surgery respectively. The pathological changes of renal tissue were observed under light microscope. The expression of TGF-β1 and BMP-7 in tubulointerstitium was detected by immunohistochemistry. Results Compared with normal control group, the expression of TGF-β1 in kidney of UUO model group was significantly increased, and the expression of BMP-7 was significantly decreased. However, after treatment with Yu Shenkang injection, the expression of TGF-β1 in UUO model rats was significantly decreased. Upregulates BMP-7 expression. Conclusion Shenkang injection can reduce renal tubulointerstitial fibrosis and delay the progression of chronic renal failure.
