目的研究白藜芦醇(Res)预处理对心肌细胞缺血再灌注损伤的保护作用的机制。方法培养乳鼠心肌细胞,模拟I/R损伤,随机分为阴性对照组、缺血再灌组及白藜芦醇预处理组,采用激光共聚焦显微术观察Res对细胞内钙的影响;采用比色法检测心肌细胞SOD和MDA活性。结果Res预处理使得心肌细胞培养液上清中超氧化物歧化酶(SOD)活性显著升高(29.28±1.24vs22.13±1.42NU/ml,P<0.05),丙二醛(MDA)活性显著降低(8.27±1.02vs13.55±1.69μmol/L,P<0.05)。Res预处理可以显著降低细胞内钙强度,P<0.05。结论白藜芦醇通过提高心肌细胞抗氧化损伤能力及降低细胞内钙超载,而缓解由于I/R产生的氧自由基损伤和钙超载所造成的损伤,从而发挥保护作用。
Objective To study the protective mechanism of resveratrol (Res) preconditioning on myocardial ischemia-reperfusion injury. Methods The cultured neonatal rat cardiomyocytes were stimulated to simulate I/R injury and randomly divided into negative control group, ischemia reperfusion group and resveratrol pretreatment group. The effect of Res on intracellular calcium was observed by laser confocal microscopy. Colorimetric assay was used to detect the activity of SOD and MDA in cardiomyocytes. Results The activity of superoxide dismutase (SOD) in the culture supernatant of cardiomyocytes was significantly increased by Res pretreatment (29.28±1.24 vs. 22.13±1.42 NU/ml, P<0.05), and malondialdehyde (MDA) activity was significantly decreased. (8.27±1.02 vs 13.55±1.69 μmol/L, P<0.05). Res pretreatment significantly reduced intracellular calcium intensity, P<0.05. Conclusion Resveratrol can protect the cardiomyocytes from anti-oxidative damage and reduce intracellular calcium overload, and relieve the damage caused by I/R-induced oxygen free radical injury and calcium overload.
