目的应用MALDI-TOF技术,建立青霉素耐药大肠杆菌(E.coli)和敏感性大肠杆菌的差异蛋白质的信息。方法诱导青霉素耐药大肠杆菌,与敏感性大肠杆菌利用二维电泳串联质谱的蛋白质学技术,对二者差异蛋白质进行对比分析。结果建立敏感和耐青霉素E.coli差异蛋白质谱,质谱发现24个差异蛋白。耐药菌株与敏感菌株相比,7个蛋白表达下降,17个表达上升。结论 MALDI-TOF技术对于发现的差异蛋白质,筛选抗药靶标有研究意义。
Objective To establish the information of differential proteins of penicillin-resistant Escherichia coli and Escherichia coli by MALDI-TOF. Methods The penicillin-resistant Escherichia coli was induced and compared with the susceptible Escherichia coli by using two-dimensional electrophoresis tandem mass spectrometry protein technology. Results Establishment of sensitive and penicillin-resistant E. coli differential protein profiles revealed 24 differential proteins by mass spectrometry. Compared with the susceptible strains, the expression of 7 proteins decreased and 17 increased. Conclusion The MALDI-TOF technique is of great significance in screening differential proteins and screening drug-resistant targets.
