应用Box-Behnken设计-响应面法优化猴头菇粗多糖的提取工艺。以提取时间、液料比、温度为影响粗多糖提取率的主要考察因素,通过Box-Behnken试验设计,以猴头菇粗多糖提取率为响应值,应用Designexpert 7.0软件对结果进行二次多项式模型拟合,绘制等高线图和响应面图,比较各因素交互作用对提取率影响的强弱,得出最优工艺,并对其进行验证。结果显示最佳工艺条件为:提取时间3.8 h,液料比25 m L/g,提取温度81℃。验证实验显示猴头菇粗多糖提取率为3.09%,实测值与预测值接近。研究结果说明响应曲面法建立的模型预测性良好,能合理地优化猴头菇中粗多糖的提取工艺。
Optimization of the extraction process of Hericium erinaceus crude polysaccharides by Box-Behnken design-response surface methodology. The time, liquid-to-solid ratio and temperature were the main factors affecting the extraction rate of crude polysaccharides. Based on the Box-Behnken experimental design, the extraction rate of Hericium erinaceus crude polysaccharide was used as the response value. The results were subjected to quadratic polynomial model Fitting and drawing the contour map and the response surface map, comparing the interaction of the factors on the extraction rate of strength, the best technology, and verify. The results showed that the optimum conditions were as follows: the extraction time was 3.8 h, the liquid to material ratio was 25 m L / g, the extraction temperature was 81 ℃. Verification experiments showed Hericium crude polysaccharide extraction rate of 3.09%, the measured value and the predicted value close. The results show that the model established by the response surface method has good predictability and can reasonably optimize the extraction process of crude polysaccharides in Hericium erinaceus.
