目的通过检测不同浓度葡萄糖及葡萄糖浓度波动对单核细胞产生白介素6(IL-6)的影响,探讨血糖波动在糖尿病大血管病变发生机制中的作用。方法实验所采用的细胞为THP-1细胞株,根据葡萄糖浓度及其变化分为对照组、高葡萄糖组、葡萄糖浓度波动组和渗透压控制组。将细胞接种于六孔培养板,每12 h换液1次,留取上清液。72 h后将每次留取的上清液等量混匀后采用酶联免疫法测定IL-6浓度,比较各组差异有无统计学意义。结果高葡萄糖组IL-6浓度明显高于对照组(P=0.000),葡萄糖浓度波动组高于高葡萄糖组(P=0.000),渗透压控制组介于高葡萄糖组和葡萄糖浓度波动组之间。结论葡萄糖浓度波动较单纯高糖更能促进单核细胞分泌IL-6,提示葡萄糖浓度波动有更强的细胞毒性。
Objective To investigate the effects of different concentrations of glucose and glucose on interleukin-6 (IL-6) production in monocytes and to explore the role of blood glucose fluctuations in the pathogenesis of diabetic macroangiopathy. Methods The cells used in this experiment were THP-1 cell lines, which were divided into control group, high glucose group, glucose concentration fluctuation group and osmotic pressure control group according to the glucose concentration and its change. The cells were seeded in six-well culture plate, changing the solution every 12 h, the supernatant was collected. The concentration of IL-6 in each supernatant after 72 h was determined by enzyme-linked immunosorbent assay (ELISA). The difference between the two groups was statistically significant. Results The concentration of IL-6 in high glucose group was significantly higher than that in control group (P = 0.000), the glucose concentration was higher in fluctuating group than in high glucose group (P = 0.000), osmotic pressure control group was between high glucose group and glucose concentration fluctuating group . Conclusion The fluctuation of glucose concentration can promote the secretion of IL-6 by monocytes more than that of high glucose alone, suggesting that there is more cytotoxicity in glucose concentration fluctuation.
