目的:了解犬口服或阴道给予酮康唑后血中药物浓度。方法;建立测定大血浆中酮康唑浓度的高效液相色谱法。对血浆中酮康唑进行了定量测定。血浆样品经乙腈沉淀蛋白,上清液用氮气流吹干后重组,进样。结果:血浆中酮康唑在(0.05~20)μg/ml范围内有良好线性关系(r=0.9999),方法回收率为96.98±3,22%(n=6),日间精密度为4.7%(n=6)。结论:本法简便、灵敏;用于犬口服或阴道给予酮康唑后血药浓度测定,效果满意。
Objective: To understand the dog oral or vaginal administration of ketoconazole blood drug concentration. Methods: To establish a HPLC method for the determination of ketoconazole in large plasma. Ketoconazole in plasma was quantitatively determined. Plasma samples were precipitated by acetonitrile, the supernatant was dried by nitrogen flow and then recombined. Results: There was a good linearity (r = 0.9999) for ketoconazole in the range of (0.05-20) μg / ml. The recovery rate was 96.98 ± 3,22% (n = 6) Daytime precision was 4.7% (n = 6). Conclusion: This method is simple and sensitive; the blood concentration of ketoconazole after oral administration or vaginal administration can be measured with satisfactory results.
