我所自1970年以来,为了寻找治疗矽肺药物,共筛选700余种中草药,600余种化合物,仅有少数几种类型化合物显示疗效。如何减少盲目的广筛,发展药物设计,定向寻找高效低毒的新药一直是我们注意的课题。几年来,我们顺着一定的思路,在广筛发现小檗胺有效的基础上,找到了汉防己甲素及一系列效果更好的双苄基异喹啉生物硷,在梯络龙最初腹腔注射效果不很明显的基础上,有意识地改为灌胃给药,效果有所提高并引起了重视。本文试图对目前筛选过的具有代表性的化合物的化学结构、理化性质及其疗效的关系作一初步探讨。
Since 1970, in order to find drugs for the treatment of silicosis, we have screened over 700 kinds of Chinese herbal medicines and more than 600 kinds of compounds. Only a few types of compounds have shown efficacy. How to reduce blind screening, develop drug design, and seek new drugs with high efficacy and low toxicity have always been our focus. Over the past few years, based on certain ideas, we have found tetrandrine and a series of more effective bis-benzylisoquinoline biosaponins on the basis of the effective screening of Berbamine. On the basis that the injection effect is not very obvious, consciously changing to intragastric administration, the effect has increased and attracted attention. This article attempts to make a preliminary study of the relationship between the chemical structure, physicochemical properties, and efficacy of representative compounds currently screened.
