目的:分析红芪多糖3(HPS-3)作用于小鼠脾淋巴细胞后的差异蛋白质点,探讨红芪多糖3调节免疫的分子生物学机制。方法:提取正常小鼠脾淋巴细胞进行体外培养,用MTT法检测红芪多糖对小鼠脾淋巴细胞的刺激增殖作用,ELISA试剂盒测定培养上清中的IL-2含量,并运用双向电泳技术对脾淋巴细胞总蛋白进行分离,银染显色,PDQuest软件对凝胶图谱进行差异点分析,找出差异明显的蛋白质点。结果:HPS-3能够提高T淋巴细胞增殖能力和IL-2的分泌,实验所得2-DE凝胶图谱经PDQuest软件分析得出以下结果:HPS-3组中共有16个蛋白点发生明显改变,其中11个蛋白质点高表达,4个蛋白质点消失,1个蛋白质点低表达。结论:HPS-3能促进脾淋巴细胞增殖,显著影响小鼠脾淋巴细胞蛋白质表达。
OBJECTIVE: To analyze the differential protein spots induced by HPS-3 in splenic lymphocytes in mice and to explore the molecular mechanism of Hs-3 polysaccharide. Methods: Splenic lymphocytes from normal mice were isolated and cultured in vitro. MTT assay was used to detect the proliferation of mouse splenic lymphocytes. ELISA kit was used to determine the content of IL-2 in the culture supernatant. Two-dimensional electrophoresis The total spleen lymphocyte protein was separated and stained with silver stain. PDQuest software was used to analyze the difference of gel chromatogram and to find out the protein spots with obvious difference. Results: HPS-3 could enhance the proliferation of T lymphocytes and the secretion of IL-2. The 2-DE gel obtained from the experiment was analyzed by PDQuest software and the following results were obtained: a total of 16 protein spots in HPS-3 group changed significantly, Among them, 11 protein spots were highly expressed, 4 spots disappeared and 1 protein spot was low. Conclusion: HPS-3 can promote the proliferation of splenic lymphocytes and significantly affect the protein expression of splenic lymphocytes in mice.
