目的:为了解TNFαMAb对大鼠AHNP时ALI是否具有防治作用。方法:利用AHNP并发ALI大鼠模型,将实验大鼠随机分为三组:假手术组、AHNP组和TNFαMAb预处理组,动态观察大鼠24小时存活情况、PaO2和PaCO2、肺组织MPO活性变化、肺含水量改变以及胰、肺组织的病理改变。结果:TNFαMAb尽管没有明显改变胰腺组织的病变,但能明显降低AHNP大鼠24小时死亡率(TNFαMAb预处理组10%,AHNP组50%,P<0.05),减少肺脏PMN的聚集,减轻肺组织的损伤程度。结论:TNFα是AHNP引起ALI发生的重要炎症介质,TNFαMAb通过阻断TNFα的作用和减少肺内PMN的聚集,对AHNP时肺组织起保护作用。
Objective: To understand whether TNFαMAb has a preventive and therapeutic effect on ALI in rats with AHNP. Methods: AHNP rats were randomly divided into three groups: sham operation group, AHNP group and TNFαMAb pretreatment group. The survival of rats at 24 hours, PaO2 and PaCO2, MPO activity in lung tissue were observed , Changes in lung water content and pathological changes of pancreas and lung tissue. Results: TNFαMAb significantly reduced the 24-hour mortality rate (10% in TNFαMAb group and 50% in AHNP group, P <0.05) although it did not significantly change the pathological changes of pancreatic tissues, and decreased the PMN accumulation in the lung The degree of lung tissue damage. CONCLUSION: TNFα is an important inflammatory mediator of ALI induced by AHNP. TNFαMAb plays a protective role in lung injury induced by AHNP by blocking the effect of TNFα and reducing the aggregation of PMN in the lung.
