目的:研究一种较好的微波和足叶乙苷的联合方法,用于白血病细胞的体外净化。方法:将微波和足叶已苷分别单独以及联合使用于正常骨髓细胞,HL60,U937和K562细胞株,通过GM-CFU和tCFU的测定来评价体外净化的效果。结果:微波和足叶乙苷对GM-CFU,TCFu存活率均表现出抑制作用,抑制程度呈现出功率相关性和浓度相关性;在一定范围内,微波和足叶乙苷对3种细胞株tCFU存活率的抑制显著大于GM-CFU(P<0.05或P<0.01);微波和足叶乙苷联合作用对3种细胞株的杀伤作用比单用足叶乙苷有显著性增加(P<0.05),而不显著增加对GM-CFU的抑制(P>0.05)。结论:微波和足叶乙苷联合用于白血病细胞的体外净化能产生协同效应,值得进一步研究。
Objective: To study a better combination of microwave and etoposide for the in vitro purification of leukemia cells. METHODS: Microwave and tetrodotoside were separately and jointly used in normal bone marrow cells, HL60, U937 and K562 cell lines. The in vitro purification effect was evaluated by GM-CFU and tCFU determination. RESULTS: Microwave and etoposide inhibited the survival rate of GM-CFU and TCFu. The degree of inhibition showed the correlation between power and concentration. Within a certain range, microwave and etoposide on three cell lines were observed. The inhibition of tCFU survival rate was significantly greater than that of GM-CFU (P<0.05 or P<0.01); the combined effect of microwave and etoposide on the three cell lines was more significant than that of epinephrine alone. Increased (P<0.05) without significant increase in inhibition of GM-CFU (P>0.05). Conclusion: The combined use of microwave and etoposide for the in vitro purification of leukemia cells can produce synergistic effects and deserve further study.
