目的克隆人幽门螺杆菌 (helicobacter pylori,Hp)尿素酶 B基因 (ure B) ,并分析其核苷酸序列的特性。方法用PCR技术从临床分离的 Hp菌株基因组中扩增出 ure B基因 ,将其克隆至 p HP质粒上进行序列分析。结果克隆得到的 ure B基因长度为 1713bp,其核苷酸序列与 Gen Bank公布的序列有 6 1个碱基存在差异 ,同源为 96 .44 % ,推定的氨基酸序列同源性为99.6 5 %。结论我们所克隆的 ure B基因可用于 Hp保护性抗原 Ure B蛋白的表达并保持原有的免疫原性
Objective To clone human ure B of Helicobacter pylori (Hp) and analyze its nucleotide sequence. Methods The ure B gene was amplified from the genome of Hp isolates by PCR and cloned into p HP plasmids for sequence analysis. Results The length of ure B gene cloned was 1713bp. The nucleotide sequence of ure B gene was different from that of GenBank. The homology was 96.44% and the putative amino acid sequence homology was 99.6% . Conclusion The cloned ure B gene can be used to express the Hp protective antigen Ure B protein and maintain its original immunogenicity
