用mRNA差异显示技术从大鼠癫痫状态的海马分离了20个cDNA差异片段,对4个cD-NA片段进行序列分析并在基因库中进行相同性比较,发现ERG8、ERG11、ERG12无相同性序列,ERG14与鼠的微管相关蛋白基因有64%~69%的相同性。由于这些基因的差异表达是致痫剂马桑内酯和抗痫剂MK-801所致,所以,ERG8可能是新的候选癫痫基因,ERG11和ERG12可能是新的候选抗痫基因;由于微管相关蛋白高表达与惊厥条件下海马苔藓纤维侧枝出芽关系密切,因此,ERG14在癫痫早期高表达可能预示着新的轴突生长和突触形成。
Twenty different cDNA fragments were isolated from hippocampus of rats with epilepsy by mRNA differential display. Sequence analysis of 4 cD-NA fragments and comparison of their identities in gene pool showed that ERG8, ERG11 and ERG12 had no homology , ERG14 and mouse microtubule-associated protein gene 64% to 69% of the same. ERG8 may be a new candidate epilepsy gene, ERG11 and ERG12 may be new candidate anti-epileptogenic genes due to the differential expression of these genes is caused by the epileptogenic agent and the anti-epileptic MK-801; The high expression of related proteins is closely related to the sprouting of the lateral branches of the hippocampal moss fibers under convulsion conditions. Therefore, the high expression of ERG14 in the early epilepsy may indicate new axonal growth and synapse formation.
