目的 研究脂多糖 (LPS)刺激PCP大鼠肺泡巨噬细胞后C/EBPε、IL - 1、IL - 6、TNFα的mRNA表达和相互调变关系及其对PCP大鼠肺泡巨噬细胞的免疫调节作用。方法 取PCP大鼠的肺泡巨噬细胞 ,加入LPS培养 ,分别于 1h、4h、8h及2 4h取贴壁细胞 ,提取总RNA ,用半定量RT -PCR检测C/EBPε、IL - 6、TNFα的mRNA表达。结果 我们发现 ,PCP大鼠肺泡巨噬细胞在LPS的诱导下 ,TNFα和IL - 6的表达在 1~ 2 4h内表达均明显增强 (p <0 0 5 ) ,C/EBPε的表达呈逐渐上升趋势 ,8h后可见高表达 (p <0 0 5 ) ,C/EBPε达到高峰的时间较TNFα和IL - 6晚。 结论 PCP大鼠肺泡巨噬细胞在LPS的诱导下 ,产生具有免疫活性的细胞因子 (如IL - 6、TNFα等 )明显增加 ,这些细胞因子作为生物信息正反馈回上游基因 ,进一步引起C/EBPε表达增强。
Objective To investigate the relationship between mRNA and protein expression of C / EBPε, IL - 1, IL - 6 and TNFα in alveolar macrophages stimulated by lipopolysaccharide (LPS) and their immunomodulatory effects on alveolar macrophages of PCP rats effect. Methods Alveolar macrophages of PCP rats were obtained and cultured in LPS. Adherent cells were harvested at 1h, 4h, 8h and 24h, respectively. Total RNA was extracted and C / EBPε, IL - 6 and TNFα MRNA expression. Results We found that the expression of TNFα and IL - 6 in PCP rat alveolar macrophages induced by LPS were significantly increased in 1 ~ 24 h (p <0.05), and the expression of C / EBPε was gradually increased (P <0 05). The C / EBPε peaked later than that of TNFα and IL - 6. Conclusions The alveolar macrophages in PCP rats induced a significant increase in cytokines (such as IL - 6 and TNFα) induced by LPS. These cytokines were positively and inversely fed back to the upstream genes as bioinformatics, further causing C / EBPε Enhanced expression.
