[目的]探讨三氧化二砷(As2O3)对人肝癌细胞生长抑制和诱导凋亡作用及其对细胞内活性氧(ROS)水平的影响。[方法]用MTT法、DNA Ladder检测及流式细胞术等观察As2O3对人肝癌细胞株HepG2的生长抑制和诱导凋亡作用及细胞内ROS的变化。[结果]As2O3能明显抑制HepG2细胞的增殖,并呈时间和剂量依赖性。DNA Ladder检测示,12μmol/LAs2O3处理HepG2细胞48h开始出现DNA Ladder条带。流式细胞仪分析显示As2O3处理人肝癌细胞HepG212、24、36h后细胞内ROS水平较对照组明显升高(P<0.01),且呈时间依赖性。[结论]As2O3可通过抑制人肝癌细胞HepG2增殖和提高细胞内ROS水平诱导细胞凋亡而发挥抗肿瘤作用。
[Objective] To investigate the effect of As2O3 on the growth inhibition and apoptosis induction of human hepatocarcinoma cells and its effect on intracellular reactive oxygen species (ROS). [Methods] MTT assay, DNA Ladder assay and flow cytometry were used to observe the growth inhibition, apoptosis induction and intracellular ROS of As 2 O 3 on human hepatocellular carcinoma cell line HepG2. [Results] As2O3 significantly inhibited the proliferation of HepG2 cells in a time and dose-dependent manner. DNA Ladder test showed that DNA Ladder bands appeared in HepG2 cells treated with 12μmol / L As2O3 for 48h. Flow cytometry analysis showed that the intracellular ROS level of HepG212 cells treated with As2O3 for 24, 36 and 36 hours was significantly higher than that of the control group (P <0.01), and in a time-dependent manner. [Conclusion] As2O3 can exert antitumor effect by inhibiting the proliferation of HepG2 cells and increasing the level of intracellular ROS.
