目的探讨一维及三维纳米TiO2和常规TiO2的遗传毒性。方法人离体淋巴细胞培养,采用不同剂量三种不同材料的TiO2染毒,通过观察淋巴细胞转化率,并结合单细胞凝胶电泳实验、微核实验测定细胞DNA损伤。结果淋巴细胞转化率各染毒组与阴性对照差异为无统计学意义;单细胞凝胶电泳实验显示1D TiO2拖尾率与常规TiO2相比差异无统计学意义,3D TiO2与常规TiO2之间的差异有统计学意义(P<0.05);TiO2染毒各个剂量组的微核率与阴性对照差异均无统计学意义。结论3D TiO2具有一定的遗传毒性,而1D TiO2表现不明显。
Objective To investigate the genotoxicity of one-dimensional and three-dimensional nano-TiO2 and conventional TiO2. Methods Human lymphocytes were cultured in vitro. The cells were exposed to different doses of TiO2. The lymphocyte transformation rate was observed. Single cell gel electrophoresis (PAGE) and micronucleus test were used to determine DNA damage. Results There was no significant difference in the rate of lymphocyte transformation between the treated and the negative control groups. The single cell gel electrophoresis showed that there was no significant difference between the tailing rate of 1D TiO2 and that of conventional TiO2. The differences were statistically significant (P <0.05). There was no significant difference between the micronucleus rate and the negative control in each dose of TiO2 exposure. Conclusion 3D TiO2 has a certain degree of genotoxicity, while the 1D TiO2 shows no obvious effect.
