〔目的〕研究适用于国境卫生检疫对输入性蚊类携带的登革病毒检测的方法。〔方法〕针对国境口岸输入性蚊类样本的特点和检疫检测的要求,筛选与试验对蚊样本检测登革病毒快速、敏感、特异的技术和方法。〔结果〕口岸输入性蚊类样本具有蚊虫数量少、不可重复获得等特点,需要有检测小样本蚊虫的方法,大样本检测需要一次完成并结果较准确。〔结论〕确定了适合对蚊虫小样本检测登革病毒的方法。“单克隆抗体免疫荧光法”和“RT-PCR”技术被用于对蚊虫大样本进行病毒分离及鉴定,并制定了相应的行业标准规程。
[Objective] To study the method applied to the detection of dengue virus carried by imported mosquitoes in frontier health quarantine. [Methods] According to the characteristics of imported mosquito samples at border crossings and quarantine inspection requirements, the screening and testing of rapid, sensitive and specific techniques and methods for detecting dengue virus in mosquito samples were carried out. [Result] The imported mosquito samples at ports have the characteristics of fewer mosquitoes, no repeated acquisition and so on. Therefore, the method of detecting small samples of mosquitoes is required. The detection of large samples needs to be completed once and the result is more accurate. [Conclusion] A suitable method for detecting dengue virus in a small sample of mosquitoes was identified. The “monoclonal antibody immunofluorescence” and “RT-PCR” techniques were used to isolate and identify large mosquito samples and to develop corresponding industry standard protocols.
